THE SCIENCE.
PARKINSON’S DISEASE.
Parkinson's Disease (PD) is characterized by the death of cells (neurons) in many areas of the brain and throughout the body. Programmed cell death is initiated by lack of NAD+ and NAD+ is a coenzyme found within every cell to help drive enzymatic reactions forward.
Mitochondrial dysfunction, combined with low levels of NAD+, can significantly alter a cell’s ability to produce energy and maintain proper function. Neurons are especially susceptible to energy fluctuations due to mitochondrial dysfunction. If neurons become damaged, it can lead to several neurodegenerative diseases. NAD+ also regulates a class of proteins known as sirtuins. Sirtuins regulate anti-inflammatory processes throughout the body and the brain and any sirtuin implications can lead to some neurodegenerative diseases. An increased production of NAD+ helps to increase the expression of sirtuins and promote neurogenesis (vital for the loss of cells involved with Parkinson's Disease). PARP, also known as polymerases, also play a key role in DNA repair and are crucial for cell health. Hyper-activation of PARP (which occurs for those with Parkinson's Disease) has been shown to deplete NAD+.
All-in-all, mitochondrial dysfunction, sirtuin implications and the hyper-activation of PARP all directly affect the cells in many areas of the brain and body, leading to cell death and lower levels of NAD+.
According to NADTreatmentCenter.com, Prescription drugs only help reduce the symptoms of PD. There is no widely accepted preventative measure that treats the underlying pathology for PD to date. Every year 60,000 new people are diagnosed with PD and $25 billion is spent to treat PD annually. The pressure for effective preventative measures and curative treatments for PD is rising with each new diagnosis.
ALZHEIMER’S DISEASE.
The precise cause of Alzheimer’s Disease is unknown, but it is characterized by the death of brain cells. In Alzheimer’s Disease, the brain’s usual DNA repair activity is impaired, leading to mitochondrial dysfunction, lower neuron production, and increased neuronal dysfunction and inflammation. NAD+ is a compound that the brain needs to regulate DNA repair and other key signaling pathways. As NAD+ levels decline, the brain cells lose a critical DNA repair function that appears to inflict neurodegenerative diseases such as Alzheimer’s Disease.
Neuroscientists of the Johns Hopkins University School of Medicine teamed up with researchers at the National Institute on Aging to identify core mechanisms that contribute to Alzheimer’s disease development.
Their findings that were published in the Proceedings of the National Academy of Science, show that boosting NAD+ beneficially alters some of the pathological processes that contribute to brain-cell death in Alzheimer’s disease.
